ABSTRACT

There are several reports on herbicide paraquat (PQ)-induced Parkinsonian-like pathology in different animal models, including Drosophila melanogaster. Also, the role of some inflammatory factors, such as nitric oxide is reported in PQ-induced neuroinflammation of Drosophila. Although invertebrate model is valuable to study the conserved inflammatory pathway at the time of neurodegeneration, but neuroinflammation during PQ-mediated neurodegeneration has not been studied explicitly in Drosophila. In this study, the inflammatory response was examined in Drosophila model during PQ-induced neurodegeneration. We found that after exposure to PQ, survivability and locomotion ability were affected in both sexes of Drosophila. Behavioural symptoms indicated similar physiological features of Parkinson’s disease (PD) in different animal models, as well as in humans. Our study revealed alteration in proinflamatory factor, TNF-α and Eiger (the Drosophila homologue in TNF superfamily) was changed in PQ-treated Drosophila both at protein and mRNA level during neurodegeneration. To ensure the occurrence of neurodegeneration, tyrosine hydroxylase (TH) positive neuronal cell loss was considered as a hallmark of PD in the fly brain. Thus, our result revealed the conserved inflammatory events in terms of expression of TNF-α and Eiger present during a sublethal dose of PQ-administered neurodegeneration in male and female Drosophila with significant variation in proinflamatory factor level among both the sexes.

ABSTRACT

La esclerosis múltiple es una enfermedad inflamatoria desmielinizante que afecta el sistema nervioso central y que es considerada una de las principales causas de discapacidad en jóvenes adultos. Las causas de la esclerosis múltiple son aún desconocidas, aunque se cree que una combinación de factores genéticos y ambientales resulta en una respuesta autoinmune que promueve la degeneración neuronal/axonal. En esta revisión se analiza la asociación entre la respuesta inmune y la neurodegeneración en la esclerosis múltiple.


Multiple sclerosis is an inflammatory demyelinating disease affecting the central nervous system and considered one of the leading causes of disability in young adults. The precise cause of multiple sclerosis is unknown, although the current evidence points towards a combination of genetic and environmental factors leading to an autoimmune response that promotes neuronal degeneration. In this review, we will describe the association between the immune response and neurodegeneration in multiple sclerosis.

ABSTRACT

La infección de tejidos cerebrales por HIV se asocia a desórdenes neurocognitivos identificados bajo la denominación HAND y categorizados en demencia, formas moderadas y formas asintomáticas. La introducción de terapia de alta efectividad ha implicado un notorio descenso de la demencia, pero no ha influido sobre las formas moderadas y asintomáticas que afectan alrededor del 50% de los pacientes bajo tratamiento. Esa disfunción cognitiva resulta de la pérdida de neuronas que, sin embargo, no han sido infectadas por el virus. De ahí la importancia de mecanismos indirectos en la neuropatogenia de HIV, ya que las citocinas/quimiocinas pro-inflamatorias liberadas por macrófagos/microglia infectados, la excitoxidad y el stress oxidativo se evidencian como principales causas de injuria neuronal, además de la directamente provocada por proteínas virales. Un mejor conocimiento de la interacción de HIV con su huésped humano está posibilitando el desarrollo de abordajes diganósticos más confiables y de estrategias terapéuticas más efectivas a nivel de SNC


HIV-1 associated neurocognitive disorders (HAND) result from brain infection. They are categorized as dementia, mild cases and asymptomatic cases. The introduction of HAART has markedly decreased dementia but no influence has been observed in mild and asymptomatic cases, since they are still identified in around 50 % of treated patients. Such cognitive dysfunction is the outcome of the loss of neurons which, however, have not been infected by the virus. hence, the importance of indirect mechanisms in HIV neuropathogenesis in which cytokines/chemokines released by infected macrophages/microglia, excitotoxic neuronal injury and oxidative stress are relevant causes of neurodegeneration besides that exerted by viral proteins. A better understanding of the HIV interaction with th human host is enabling the development of more reliable diagnostic biomarkers and more effective therapeutic strategies at CNS level

ABSTRACT

O prion celular (PrPc) é uma glicoproteína ligada à membrana plasmática por uma âncora de GPI (glycosylphosphatidylinositol). A sua isoforma anormal (PrPsc) é uma molécula infecciosa que causa várias doenças neurodegenerativas em mamíferos. A etiologia dessas doenças está associada a uma mudança conformacional pós-traducional de PrPc que ocorre após sua internalização (Prusiner), 1998. Na tentativa de desvendar as funções fisiológicas de PrPc, nosso grupo tem identificado e caracterizado as interações celulares que PrPc participa. A primeira delas é a interação entre PrPc e STI1 (Stress Inducible Protein 1). Essa interação transduz sinalização por cAMP e PKA levando a neuroproteção contra morte celular programada(Chiarini e cols.2002; Zanata e cols, 2002)

ABSTRACT

Os efeitos benéficos do extrato padronizado de Ginkgo bilola (EGb 761) em doenças neurodegenerativas parecem estar relacionados com alterações da circulação cerebral e/ou com sua atividade antioxidante. Porém, os mecanismos envolvidos nestas atividades não estão completamente estabelecidos. Neste trabalho avaliou-se a atividade das enzimas catalase, superóxido dismutase e gulationa peroxidase nas estruturas cerebrais hipocampo, estriado e substância negra de ratos tratados com EGb e verificou-se a ação do extrato sobre a lipoperoxidação em hipocampo. ABSTRACT

Na doenca de Alzheimer (DA), os principais eventos associados a neurodegeneracao sao a formacao de placas senis e de emaranhados neurofibrilares. Estes fenomenos relacionam-se respectivamente a deposicao de beta-amiloide (Ab) e a alteracoes do estado de fosforilacao da proteina Tau. Esta e componente essencial dos microtubulos, onde se encontra em estado polimerizado. A estabilidade do polimero depende do grau de fosforilacao da Tau, tornando-se mais instavel quanto mais fosforilada a proteina. Consequentemente, a hiperfosforilacao da Tau relaciona-se com menor estabilidade do citoesqueleto, favorecendo a morte neuronal. O Ab e produzido pela clivagem da proteina precursora do amiloide (APP) por acao da enzima beta-secretase, em detrimento da acao mais fisiologica da alfa-secretase, que da origem ao fragmento APPs. As fibras de Ab tem diversos efeitos neurotoxicos, alem de ocorrerem associadamente a uma presumivel perda funcional do metabolito secretado APPs...
